Can't connect

Hi Steve i have been trying to set up my profilux system and i'm using the rs232 - usb connection.

Now i have loaded the drivers for the cable and it is showing in the device manager as comm 12
I have changed the comm port on the programme page the three dots bit as it has stated in your instructions,but soon as i press connect it's saying no connection

Do i have to have everything plugged in to get it working or can i just set it up with just the controller and my pc
